The relative importance of multiple vectors to the initial establishment, spread and population dynamics of invasive species remains poorly understood. This study used molecular methods to clarify the roles of commercial shipping and recreational boating in the invasion by the cosmopolitan tunicate, Botryllus schlosseri. We evaluated (i) single vs. multiple introduction scenarios, (ii) the relative importance of shipping and boating to primary introductions, (iii) the interaction between these vectors for spread (i.e. the presence of a shipping-boating network) and (iv) the role of boating in determining population similarity. Tunicates were sampled from 26 populations along the Nova Scotia, Canada, coast that were exposed to either shipping (i.e. ports) or boating (i.e. marinas) activities. A total of 874 individuals (c. 30 per population) from five ports and 21 marinas was collected and analysed using both mitochondrial cytochrome c oxidase subunit I gene (COI) and 10 nuclear microsatellite markers. The geographical location of multiple hotspot populations indicates that multiple invasions have occurred in Nova Scotia. A loss of genetic diversity from port to marina populations suggests a stronger influence of ships than recreational boats on primary coastal introductions. Population genetic similarity analysis reveals a dependence of marina populations on those that had been previously established in ports. Empirical data on marina connectivity because of boating better explains patterns in population similarities than does natural spread. We conclude that frequent primary introductions arise by ships and that secondary spread occurs gradually thereafter around individual ports, facilitated by recreational boating.  相似文献

Wheat-spikes inoculated withN. indica at the boot-leaf stage produced secondary sporidia when later incubated (intact/detached) under moist conditions in the laboratory. Sporidia were also released from inoculated spikes in the field where sporidial release exhibited diurnal periodicity. More sporidia were trapped between 5–6 o'clock than during the later parts of the day but no sporidia were trapped between 14–18 o'clock. However, they could be trapped at any time of the day from the detached spikes incubated under moist conditions in the laboratory. Sporidia trapped in different experiments were invariably of the allantoid type and they proved viable and infective. Maximum sporidia developed on the outer glumes of florets, and this observation was supported by scanning electron microcope studies. Sporidia developed at 15 and 20°C but not at 30°C. These findings indicated that repeated cycles of sporidial production in spikes provided more inoculum than expected from soil-borne teliospores ofN. indica.  相似文献

ABSTRACT. Spores of Nosema bombycis Y9101, isolated from the beet armyworm, Spodoptera exigua , were primed with an alkaline solution and inoculated into Antheraea eucalypti cell cultures. Infected cells were subcultured every five days at three cell densities (2.5 × 103, 5.0 × 103, and 1.0 × 104 cells/cm2). A difference was observed in the spread of N. bombycis Y9101 infection between low-density and higher-density cultures of host cells. The host cell density did not affect the productivity of secondary infective forms of the parasite. The principal factor determining the rate of microsporidian infection in vitro was the number of host cells existing within the reach of extruded short-coiled polar tubes from spores germinated intracellularly.  相似文献

When mechanically inoculated to susceptible tobacco (Nicotiana tabacum L.) cultivars, nine isolates of PVY from Umbria (Central Italy) and two from Southern Latium gave rise to rapid systemic infection which developed within 6–8 days after inoculation. Systemic spread of the same isolates was slower, or much slower, in infected pepper (Capsicum annuum L.) cultivars, 8–14 days for Southern Latium isolates and 20 - 35 days for Umbrian ones. Aphid (Myzus persicae)-moculation of pepper and tobacco plants with two of the Umbrian and one of the Southern Latium isolates confirmed the results from sap-transmission and showed that fewer inoculated pepper plants become infected, especially with Umbrian isolates. In agreement with the data on systemic spread, aphid-acquisition trials indicated that tobacco plants became efficient PVY sources for vectors 6–8 days after inoculation with either group of isolates. Peppers became efficient acquisition hosts 8–15 days after inoculation with Southern Latium isolates but not until 22–45 days after inoculation with Umbrian ones. Southern Latium isolates induced more severe symptoms in pepper cultivars than Umbrian isolates did. One of the Southern Latium isolates was able to systemically infect the resistant pepper cv. Yolo Y, which was never infected by the Umbrian isolates. The Umbrian isolates tested seem to be better adapted to tobacco than peppers, while Southern Latium ones are well adapted to both.  相似文献

Summary Intact and denervated extensor digitorum longus (EDL) muscles of 20-day-old inbred Lewis-Wistar rats were labelled with 3H-thymidine. Ninety minutes after the injection of the isotope 4.0% of the nuclei were labelled in the intact (i.e. innervated) and 9.6% in the muscles, denervated 3 days before administration of the isotope. The labelled EDL muscles were grafted into the bed of the previously removed EDL muscles of inbred animals and these isografts were studied 30 days later.In the EDL muscles, regenerated from innervated isografts only occasionally labelled endothelial cells were found whereas in the muscles regenerated from denervated isografts also parenchymal muscle nuclei were regularly labelled. The incidence of labelled nuclei in the regenerated EDL muscles was, however, about 20 times lower than in the donor EDL muscles. The present experiments provide a direct proof of utilization of donor satellite cell nuclei for regeneration in grafted muscle tissue. With respect to the low incidence of labelled nuclei in regenerated EDL muscles, other sources of cells apparently also contribute to the regeneration process.  相似文献

Prions are misfolded infectious proteins responsible for a group of fatal neurodegenerative diseases termed transmissible spongiform encephalopathy or prion diseases. Chronic Wasting Disease (CWD) is the prion disease with the highest spillover potential, affecting at least seven Cervidae (deer) species. The zoonotic potential of CWD is inconclusive and cannot be ruled out. A risk of infection for other domestic and wildlife species is also plausible. Here, we review the current status of the knowledge with respect to CWD ecology in wildlife. Our current understanding of the geographic distribution of CWD lacks spatial and temporal detail, does not consider the biogeography of infectious diseases, and is largely biased by sampling based on hunters' cooperation and funding available for each region. Limitations of the methods used for data collection suggest that the extent and prevalence of CWD in wildlife is underestimated. If the zoonotic potential of CWD is confirmed in the short term, as suggested by recent results obtained in experimental animal models, there will be limited accurate epidemiological data to inform public health. Research gaps in CWD prion ecology include the need to identify specific biological characteristics of potential CWD reservoir species that better explain susceptibility to spillover, landscape and climate configurations that are suitable for CWD transmission, and the magnitude of sampling bias in our current understanding of CWD distribution and risk. Addressing these research gaps will help anticipate novel areas and species where CWD spillover is expected, which will inform control strategies. From an ecological perspective, control strategies could include assessing restoration of natural predators of CWD reservoirs, ultrasensitive CWD detection in biotic and abiotic reservoirs, and deer density and landscape modification to reduce CWD spread and prevalence.  相似文献

Reclamation of highly disturbed lands typically includes establishing fast‐growing, non‐native plants to achieve rapid ground cover for erosion control. Establishing native plant communities could achieve ecosystem functions beyond soil erosion, such as providing wildlife habitat. Pipelines, or other disturbed corridors through a landscape, present unique challenges for establishing native plant communities given the heterogeneity of soil environments and invasive plant propagule pressure. We created two structural equation models to address multiple related hypotheses about the influence of soil pH on plant community composition (current diversity and vegetative cover of the original restoration seed mix and background flora, and invasive plant density during mix establishment and current density) of a highly disturbed landscape corridor restored with native species. To test our hypotheses we conducted a plant survey on a gas pipeline crossing two state forests in the north‐central Appalachians that had been seeded with a native‐based mixture 8 years prior. Low soil pH was a strong predictor of density of the invasive annual plant, Microstegium vimineum, and had resulted in lower species diversity and cover of the seeded mix. Overall, our data provide evidence that native‐based grass and forb mixtures can establish and persist on a wide range of soil environments and thrive in competition with invasive plants in moderately acidic to neutral soils. Advancing knowledge on restoration methods using native species is essential to improving restoration practice norms to incorporate multifunctional ecological goals.  相似文献
